Animal models are essential for basic and clinical research on virus diseases. Humanized mice (mice reconstituted with human hematopoietic cells) have been effectively used various studies as small animal models. Studies human-tropic HIV-1 also performed using different humanized mouse Various generated distinct strains engraftment methods. These techniques affect the reconstitution of cells in individual mice, turn replication vivo . In this report, we describe details generation method i.e., severely immunodeficient (NSG mice) transplanted CD133-positive via intra-bone marrow injection (IBMI). It has shown that highly capable to generate CD34-positive IBMI is an excellent methodology lymphoid myeloid cell repopulation. into bone marrow, lymphocytes were increased 3 months after transplantation a steady increase CD4-positive was observed until 6–8 transplantation. order test utility our system, CXCR4-tropic CCR5-tropic clones intraperitoneally inoculated resultant Upon inoculation at same dose viruses, plasma viral load HIV-1-inoculated peaked earlier than (2–3 weeks vs 5–10 post-inoculation).
